The gradual adjustment of a peptide dose — typically starting low and stepping up — to minimize side effects while establishing effective exposure.
Titration is the standard approach to compounds with dose-dependent side-effect profiles — most notably GLP-1 analogs, where starting at therapeutic dose produces unmanageable nausea in most researchers. The established Semaglutide titration starts at 0.25mg weekly, steps to 0.5mg at week 5, then 1mg, 1.7mg, 2.4mg at 4-week intervals. This pattern is deliberately slow because receptor adaptation takes time.
Titration also applies in reverse (de-titration) when discontinuing a compound. Abrupt discontinuation of some peptides produces rebound effects; stepped reduction allows downstream pathways to re-equilibrate.
