Gonadorelin

GnRH / Gonadotropin-Releasing Hormone / Factrel / Lutrepulse

A synthetic decapeptide identical to endogenous gonadotropin-releasing hormone (GnRH). When administered in pulsatile fashion, it stimulates the pituitary to release LH and FSH, preserving testicular function and fertility. Increasingly prescribed alongside TRT to maintain intratesticular testosterone and spermatogenesis — replacing HCG for many patients after the 503A compounding reclassification.

Evidence Overview

Strength of evidence across clinical, expert, and experimental lanes.

How Gonadorelin is dosed in practice

Usually taken by subcutaneous (SubQ) injection. Typical range: 100–400 mcg 2x/week. Half-life: 2–4 minutes. Each card gives the dose, frequency and duration from one evidence lane: Clinical (human trials), Expert (practitioner protocols) or Experimental (early or animal data). These ranges describe what has been studied or used; they are not a prescription.

Work out the units to draw with the reconstitution calculator, how long a vial lasts with the vial duration calculator, and how levels fall between doses with the half-life calculator.

clinical55% confidence

The human gonadorelin product (a single 100 mcg diagnostic pituitary test) has been discontinued in the US. Human trials used gonadorelin in a pulsatile pump for men with hypogonadotropic hypogonadism, not as twice-weekly injections; no published trial has tested it alongside TRT.

Dose100 mcg single diagnostic dose (SubQ or IV); about 25 ng/kg per pulse by pump for hypogonadotropic hypogonadism
FrequencyOnce (diagnostic); a pump pulse every 90 minutes (fertility treatment)
DurationSingle test; months of pump therapy

Now the standard replacement for HCG in TRT protocols after compounding pharmacy restrictions. 100–200 mcg SubQ twice weekly preserves testicular size and spermatogenesis. Some clinics use 3x/week for aggressive fertility preservation. Monitor LH, FSH, and estradiol. Much shorter half-life than HCG requires more precise dosing.

Dose100–200 mcg 2–3x weekly
Frequency2–3x weekly SubQ
DurationOngoing alongside TRT

Gonadorelin: common questions

What is Gonadorelin?

A synthetic decapeptide identical to endogenous gonadotropin-releasing hormone (GnRH). When administered in pulsatile fashion, it stimulates the pituitary to release LH and FSH, preserving testicular function and fertility. Increasingly prescribed alongside TRT to maintain intratesticular testosterone and spermatogenesis — replacing HCG for many patients after the 503A compounding reclassification.

How is Gonadorelin dosed?

Gonadorelin is usually taken by subcutaneous (SubQ) injection. Typical range in DoseCraft's library: 100–400 mcg 2x/week. Clinical (human trials): 100 mcg single diagnostic dose (SubQ or IV); about 25 ng/kg per pulse by pump for hypogonadotropic hypogonadism; Once (diagnostic); a pump pulse every 90 minutes (fertility treatment); Single test; months of pump therapy. Expert (practitioner protocols): 100–200 mcg 2–3x weekly; 2–3x weekly SubQ; Ongoing alongside TRT. These ranges describe what has been studied or used; they are not a prescription.

Is Gonadorelin FDA-approved?

Gonadorelin is not an FDA-approved finished drug. DoseCraft's library lists it as prepared by compounding pharmacies, which depends on current FDA rules for that substance.

What is the half-life of Gonadorelin?

DoseCraft's library lists a half-life of 2–4 minutes. Half-life sets how often a compound is dosed and how long it takes to clear: about five half-lives.

Who should not use Gonadorelin?

Listed contraindications: GnRH-dependent tumors (prostate cancer); Known hypersensitivity to GnRH analogs; Pregnancy (teratogenic risk).
